:= is an assignment operator in PL/SQL. It's purpose is to assign a value to some variable.
(Do not confuse this with the equal to sign = Eh!)
eg.
vAvgAge := 32;
vMyName := 'TOM';
IF vMyDesignation = NULL THEN
vMyDesignation := vDefaultDesignation;
END IF;
